Preparation
Mac suspension: Each 5 ml of reconstituted suspension contains erythromycin ethylsuccinate USPequivalent to 125 mg of erythromycin.
Mac DS suspension: Each 5 ml of reconstituted suspensioncontains erythromycin ethylsuccinate USP equivalent to 250 mg of erythromycin.
Indication
Upper respiratory tract infections: Tonsillitis, Pharyngitis, Laryngitis, Sinusitis, Influenza.
Lower respiratory tract infections: Acute and chronic bronchitis, Pneumonia.
Oral infections: Gingivitis.
Skin & soft tissue infections: Boils & Carbuncles, Abscesses, Impetigo, Cellulitis.
Gastrointestinal infections: Acute Gastroenteritis, Cholecystitis. Prophylaxis: Rheumatic fever, Endocarditis, Pre &Post operative trauma.
Other infections: Osteomyelitis, Urethritis, Gonorrhoea, Syphilis,Diphtheria.
Contraindication
It is contraindicated in patients hypersensitive to erythromycin. Usage In pregnancy & lactationperiod Caution should be exercised when prescribing the drug for the pregnant women &lactating women.Directions for reconstitutionMac 100 ml suspension: Shake the bottle to loosen powder. Add 85 ml (approx. 17 measuringspoonful) of boiled and cooled water to the dry powder in the bottle. For ease of preparation, addwater to the bottle in two portions. Shake well after each addition until all the powder is insuspension.Mac DS 100 ml suspension: Shake the bottle to loosen powder. Add 70 ml of boiled and cooledwater to the dry powder in the bottle. For ease of preparation, add water to the bottle in twoportions. Shake well after each addition until all the powder is in suspension. Mac DS 70 mlsuspension: Shake the bottle to loosen powder. Add 50 ml of boiled and cooled water to the drypowder in the bottle. For ease of preparation, add water
Dosage & Administration
Infants and babies up to 2 years: 125 mg every six hourly.
Children aged 2-8 years: 250 mg everysix hourly.
Children aged over 8 years & adults: 500 mg every six hourly. For severe infections the recommended dose may be doubled.
Acute Gastroenteritis: 30-50 mg/kg/day orally in q.i.d. for 5days. Pertussis: 40-50 mg/kg/day given in divided doses.
Prophylaxis of endocarditis: 10 mg/kgorally two hours before to the procedure and then 5 mg/kg/ six hours after the initial dose.
Side Effect
The most frequent side effects are gastrointestinal which include nausea, vomiting, abdominalpain, diarrhoea and anorexia. There have been isolate reports of transient CNS dysfunction.Occasional reports of cardiac arrhythmia have been documented.
Precaution
Keep the bottle tightly closed and protect from light. Store in a cool and dry place. Bottle shouldbe shaken well before use.
Drug Interaction
Erythromycin has the potential to interact with many medications by inhibiting drug metabolism in the liver. Several reports and controlled studies have shown that erythromycin may interact with theophylline, carbamazepine, cyclosporin, tacrolimus, warfarin, digoxin, terfenadine, astemazole, cisapride, lovastatin, triazolam, and disopyramide.
